چکیده

The clinical demand for cartilage tissue engineering is potentially large for reconstruction defects resulting from congenital deformities or degenerative disease due to limited donor sites for autologous tissue and donor site morbidities. Cartilage tissue engineering has been successfully applied to the medical field: a scaffold pre-cultured with chondrocytes was used prior to implantation in an animal model. We have developed a surgical approach in which tissues are engineered by implantation with a vascular pedicle as an in vivo bioreactor in bone and adipose tissue engineering. Collagen type II, chitosan, poly(lactic-co-glycolic acid) (PLGA) and polycaprolactone (PCL) were four commonly applied scaffolds in cartilage tissue engineering. To expand the application of the same animal model in cartilage tissue engineering, these four scaffolds were selected and compared for their ability to generate cartilage with chondrocytes in the same model with an in vivo bioreactor. Gene expression and immunohistochemistry staining methods were used to evaluate the chondrogenesis and osteogenesis of specimens. The result showed that the PLGA and PCL scaffolds exhibited better chondrogenesis than chitosan and type II collagen in the in vivo bioreactor. Among these four scaffolds, the PCL scaffold presented the most significant result of chondrogenesis embedded around the vascular pedicle in the long-term culture incubation phase.

Comparison between Chondrogenic Markers of Differentiated Chondrocytes from Adipose Derived Stem Cells and Articular Chondrocytes In Vitro

Objective(s): Osteoarthritis is one of the most common diseases in middle-aged population in the world. Cartilage tissue engineering (TE) has been presented as an effort to introduce the best combination of cells, biomaterial scaffolds and stimulating growth factors to produce a cartilage tissue similar to the natural articular cartilage. In this study, the chondrogenic potential of adipose d...

مقایسه توانایی ایجاد محیط رشد و تمایز مناسب سلول‌های بنیادی مزانشیمی مشتق از بافت چربی انسانی بین داربست‌های زیستی مختلف

Background: Although the presence of articular cartilage progenitor cells is already known, but due to lack of blood supply, its ability to heal is poor. In past, tissue engineering approaches has been mainly focused on the collection and culture of chondrocyte cells on biodegradable scaffolds such as polyglycolic acid in vitro and their autologous transplantation. Recently, due to the difficul...

Comparison of the efficacy of Piascledine and transforming growth factor β1 on chondrogenic differentiation of human adipose-derived stem cells in fibrin and fibrin-alginate scaffolds

Objective(s):The aim of this study was to compare the chondrogenic induction potential of Piascledine and TGF-β1 on adipose-derived stem cells (ADSCs) in fibrin and fibrin-alginate scaffolds.  Materials and Methods: Human subcutaneous adipose tissues were harvested from three patients who were scheduled to undergo liposuction. Isolated ADSCs were proliferated in a culture medium. Then, the cell...

تمایز سلول های بنیادی مزانشیمی مشتق از بافت چربی انسانی به سلول های غضروفی با استفاده از داربست هیدروژل فیبرینی

Background and purpose: Mesenchymal stem cells (MSCs) are the most widely used cell sources for cartilage tissue engineering. In the present study, human stem cells were used as a cell source. Scaffolds play an important role in tissue engineering, therefore, this study aimed at evaluating the ability of fibrin scaffolds in chondrogenic differentiation of adipose-derived mesenchymal stem cells ...

Biological and MRI characterization of biomimetic ECM scaffolds for cartilage tissue regeneration.

Osteoarthritis is the most common joint disorder affecting millions of people. Most scaffolds developed for cartilage regeneration fail due to vascularization and matrix mineralization. In this study we present a chondrogenic extracellular matrix (ECM) incorporated collagen/chitosan scaffold (chondrogenic ECM scaffold) for potential use in cartilage regenerative therapy. Biochemical characteriz...
